Research Project
Antitumour Platinum compounds on neural cell lines. Microscopical and biomolecular analysis
The research activity is addressed to the study of cellular and molecular effects induced by cisplatin and a new platinum compound, the [Pt(O,O'-acac)(γ-acac)(DMS)], on human and rat neuroblastoma and glioblastoma cell lines. In particular relevance will be given to the study of cellular accumulation of the two compounds, new cellular pathways triggered, effects on intracellular calcium homeostasis, interaction with the blood-brain barrier and epigenetic mechanisms involved in drug-resistance (miRNAs).
PtAcacDMS is stable under physiological conditions, acts on the regulation of intracellular calcium and, compared to platinum, it displays a low neurotoxicity in normal tissues and a low reactivity with DNA, suggesting a possible interaction with non-genomic targets, such as proteins involved in key pathways of cell metabolism.
