Research project

Molecular methods applied to the histopathological diagnosis of cancers: from oncogenesis to predictive factors

Mutational analysis of oncogenes implicated in Langerhans Cell Histiocytosis
The purpose of my PhD in Genetics, Molecular and Cellular Biology is to investigate the mutational status of some oncogene in the Histiocytosis.
Histiocytoses encompass a wide range of rare and heterogeneous diseases characterized by the accumulation and proliferation of histiocytes within various tissues. In particular, my study is based on Langerhans Cell Histiocytosis or LCH. The etiology of LCH is unknown. This pathology is a rare neoplasm that occurs predominantly in male children, the incidence of LCH is 2-5 per million children per year in Western Europe.
We have selected a number of cases that arrived to the attention of the Anatomic Pathology Section, University of Pavia. I will study some oncogene implicated in MAPK and PI3K/AKT pathways like BRAF, KRAS, EGFR, c-KIT, PDGFRα, PIK3CA.
The identification of possible mutations in these oncogene could represent a new therapeutic approach for LCH patients. In fact there are inhibitor drugs for this genetic alteration (target therapy).
