TECHNICAL SKILLS

Microbiological techniques: Preparation of solid and liquid culture media; techniques of sterilization (autoclave, oven, UV); preparation of bacterial cultures (E. coli, S. coelicolor, M. smegmatis); methods for determining the antimicrobial activity of different antibiotics; preparation of competent cells and transformation of E. coli and M. smegmatis using electroporation.
Molecular biology techniques: extraction of plasmidic DNA; extraction of genomic DNA from enteric bacteria and mycobacteria; extraction of total RNA from mycobacteria; DNA digestion with restriction enzymes; cloning techniques; PCR amplification; Southern blotting; Redirect-technology; construction of a genomic library; Retrotranscription-PCR, Real-Time PCR using: intercalating dyes (e.g. SYBR Green) and probes specific for a fragment (e.g. Dual-labeled probes, TaqMan assay).
Bioinformatics techniques: molecular phylogenetic analysis within the genus Mycobacterium, multiple sequence alignment, identification of orthologous genes and evaluation of the degree of homology.
