Visit to Seed Companies in Netherlands (“Seed Valley”). The visit, organized and supported by Eng. Jan Hoopman (Hoopman Group, Aalten) and Dr. Michel Kockelkoren (Agathos, Aalten), was part of the project ‘Primtech-Tecnologie Avanzate di Seed Priming per l’Industria Agro-Sementiera Lombarda’ (scientific coordinator: prof. Alma Balestrazzi).
1st day: visit to the University of Wageningen, seminar of Prof. Steven Groot, international expert in seed physiology; visit to “DEMO” Laboratories of Hoopman Group, Aalten (advanced technologies of “seed coating/drying”, “seed priming/disinfection”), seminars of Eng Jan Hoopman and Dr. Michel Kockelkoren.
2nd day: visit to Bejo Zaden BV (Trambaan), leader company in seed-horticultural branch, scientific discussion with the Dr. Bert Coompan; visit to Plantenkwekerij Gitzels BV, leader company in the horticultural branch.

Research activity concerning Ph.D. student program focused on the characterization of the two plant isoforms of the gene Tyrosyl-DNA phosphodiesterase and the chromatin remodelling in plants. Activity occurred in the Plant Biotechnology Laboratory at the Department of Biology and Biotechnology ‘L. Spallanzani’ - University of Pavia. Scientific supervisors: Prof. A. Balestrazzi and Prof. D. Carbonera.
Collaboration activity with Agro Seed Company BEJO ZADEN BV (Netherlands, www.bejo.com; Contact person Dr. Bert Compaan-R&D Department). The project is focused on the application of molecular tools adopted in the Plant Biotechnology Laboratory (Contact person Prof. Alma Balestrazzi) in relation to the seed response to genotoxic stress in samples derived from the germoplasm collection of BEJO ZADEN BV.

Research project
Analysis of Tyrosyl-DNA phosphodiesterase isoforms Tdp1α and Tdp1β and role of chromatin remodeling in plants
My research program will be focused on the molecular aspects of the genotoxic stress response in plant cells, the subject currently approached at the laboratory of Plant Biotechnology in the Department of Biology and Biotechnology of the University of Pavia, through new and integrated approaches. Plant organisms are subjected to several environmental abiotic and biotic stresses that determine the accumulation of Reactive Oxygen species (ROS) associated to deleterious effects on the genome stability. Oxidative stress fatally compromises the farm productivity and it’s necessary to define new and effective strategy with the purpose to increase the tolerance to the environmental stresses in the crop fields. New information will be acquired about the role in planta of the Tyrosyl-DNA phosphodiesterase enzymes involved in the NER and BER mechanisms, responsible of DNA repair in different DNA lesions. Functional analysis of the promoters of the genes Mt Tdp1α and Mt Tdp1β, previously isolated and characterized, will be performed through the production of transgenic lines expressing reporter gene construct. Will be so possible verify the spatio-temporal activity of the promoters and explore the possibility for biotechnological applications. In the same time a bioinformatic investigation utilizing data from RNA-seq analysis of Medicago truncatula transgenic lines silenced for the Tdp1α gene will be performed. Considering the relevant role played by the DNA repair processes, the investigation area will be extended to new model systems (ornamental species) and to new problems associated to the damage caused by ionizing radiations.
