Research project

Gene therapy experimental approaches in cellular and animal models of inherited arrhythmogenic cardiac diseases
My research is part of the studies for the characterization of Catecholaminergic Polymorphic Ventricular Tachycardia (CPVT), an inherited arrhythmic disorder that predisposes to cardiac arrest and sudden death. CPVT occurs in an autosomal dominant form caused by mutations in the gene coding for the cardiac ryanodine receptor (RyR2) and a recessive form caused by mutations in the cardiac calsequestrin (CASQ2). The recessive form of the syndrome is characterized by a decrease in calsequestrin protein expression and consequent alterations of macromolecular complexes involved in calcium homeostasis of the sarcoplasmic reticulum. The aim of my research is to develop viral constructs for gene therapy in transgenic murine models of recessive CPVT in order to obtain the recovery of an healthy phenotype. This work will include cloning and genetic engineering procedures of vectors for the gene transfer of CASQ2 and a later stage of molecular and functional characterization of animal models after infection.
